人类血小板抗原1~4系统基因分型

【Abstract】 Objective:To set up the method of genotypeing the human platelet antigen(HPA)systems from 1 through 4 by sequence-specific primers(PCR-SSP).Methods:14 primers were synthesized so that the human platelet antigen systems from 1 through 4 in 25 healthy blood donors could be genotyped by PCR-SSP method.The reusults of genotyping by PCR-SSP were compared with those obtained by allele-specific oligonucleotide hybridization(PCR-ASO) Results:The results of genotyping the 4 HPA systems by PCR-SSP were definite,satisfied and in complete agreement with those obtained by PCR-ASO method.Conclusion: The PCR-SSP method of genotyping the human platelet antigen has the advantages,such as simplicity,rapidity,accuracy and so on,opening up the broad prospects for its further application.

迄今,国际上已报道了9个血小板特异性抗原系统,包括人类血小板抗原(HPA)系统1~8以及 Naka抗原[1~3]。这些抗原都是在血小板输注和怀孕介导的同种免疫过程中被发现的。在现代临床输血工作中,为了能给新生儿同种免疫血小板减少症(NAITP)、输血后紫癜(PTP)以及血小板输注无效(PTR)等患者提供正确、及时的诊断和治疗,对病人和献血者进行血小板特异性抗原的准确分型是重要的。血清学方法是现在最普遍采用的血小板抗原分型方法,但应用一直受到抗血清来源和病人血小板数量的限制。20世纪90年代后,随着分子生物学技术的不断发展和对人类血小板抗原基因结构研究的突破性进展,使血小板基因分型更为简捷准确。笔者采用PCR序列特异引物技术(PCR-SSP)对HPA-1~4这4个比较重要的血小板抗原系统进行了基因分型研究。
1 材料与方法 1.1 样本 25名健康献血者静脉采血0.5ml,ACD抗凝。 1.2 基因组DNA抽提 采用蛋白酶K消化,酚/氯仿抽提DNA。 1.3 引物 DNA序列特异性引物设计参照文献[4,5]方法,一共12条,合成序列见表1。内对照为人类生长激素(HGH)基因特异性引物,序列: HGH-1s 5' TGCCTTCCCAACCATTGCCTTA 3';HGH-2as 5'CCACTCACGGATTTCTGTTGTGT-TTC 3';扩增产物长度为434bp。 表1 PCR-SSP引物序列、长度及其确定的特异性| 抗原系统 | 引物 | 特异性 | 碱基序列5'-3' | 长度(mer) |
